Who Should Attend?

Designed for individuals preparing genomic and/or amplicon DNA libraries for sequencing on the Ion Torrent™ systems and seeking detailed knowledge of the molecular biology techniques used for DNA library construction. Attendees will benefit by having training in a class environment, hands-on lab practicals, and interaction with fellow attendees and Ion Torrent application scientists during lecture, discussion, and lab sessions.

This class assumes attendees have prior knowledge of biological systems and basic experience with laboratory techniques such as pipetting and PCR.

Course Description:

A 1-day course involving classroom and laboratory sections for DNA library construction for sequencing on the Ion Torrent™ systems. Attendees will gain hands-on experience in constructing a DNA fragment library using control genomic DNA, and learn about different methods for shearing and size-selection, library quality assessment, amplification, quantification and troubleshooting. Class discussion also reviews amplicon library construction. (Please note, we will NOT be able to use customer samples for library construction during this course.) Information and strategies for different sequencing applications and how these relate to library construction and preparation of genomic samples will also be discussed.

Intended for first-time users of the Ion Torrent™ Personal Genome Machine™, or novice users wishing to have additional or refresher training. Attendees will benefit by having training in a class environment, hands-on lab practicals, and interaction with fellow attendees and Ion Torrent application scientists during lecture and discussion sessions.

Prerequisites:

Ownership of an Ion Torrent™ Personal Genome Machine™ (or belonging to a laboratory that has purchased a system) is a prerequisite for registration. Ideally, individuals have already participated in the live, online System and Library Training, although this is not mandatory. If this is the first training after instrument purchase, it is mandatory to have your instrument fully operational; being able to use the instrument right after the training guarantees the best use of your time. This class assumes attendees have prior knowledge of biological systems and basic experience with laboratory techniques such as pipetting and PCR.

Course Description:

An intensive overview of the Ion Torrent™ Personal Genome Machine™ technology. Course topics include an overview of instrument operation and hardware, library preparation, Ion Sphere™ particles preparation and deposition, quality control runs, troubleshooting, and data analysis. An overview of the chemistry used in the Ion Torrent™ Personal Genome Machine™ will be provided, which is critical for understanding the system and troubleshooting. Hands-on training includes initiating and monitoring sequencing runs during class, and creating Ion Sphere™ particles for template preparation using the Ion OneTouch™ System. In addition customers will be given training overview of the Ion Chef™ System, Ion Torrent’s simple to use, high throughput, modular benchtop sequencing preparation machine. Attendees will receive hands-on training which includes setup and operation of the Ion Chef™ System which automates template preparation and chip loading. Please note, we will NOT be able to use customer samples.

This two and half day course features classroom and laboratory sections for RNA library construction (small RNA and whole transcriptome). Course includes in depth discussions of RNA preparation, adapter ligation, reverse transcription, size selection, amplification, and quantification. Construction of SAGE™ libraries is not included in this course.

Course covers hands-on lab training of ultra-high multiplex PCR of targets using Ion AmpliSeq™ technology, adapter ligation, amplification, and quantification methods. Ion AmpliSeq™ Custom Primer Pools and Ion AmpliSeq™ Ready-to-use Panels are also discussed. Construction of Ion TargetSeq™ libraries is NOT included in this course.

This 2.5-day interactive course is designed for individuals who want to learn basic end-to-end workflow for aneuploidy analysis in single cells and will be directly involved in working with the Ion Reproseq™ PGS kit and the Ion Torrent™ Technology workflow. The Ion ReproSeq™ PGS kits enable the fastest benchtop next-generation sequencing workflow for aneuploidy analysis research. The course objective is to provide a comprehensive overview of the Ion ReproSeq™ PGS Kit with the Ion PGM™ System workflow, allowing users to perform detection of aneuploidies in all 24 sets of chromosomes for labs interested in rapidly and reliably researching preimplantation embryos.
